چکیده

As you will see, while the referees acknowledge that the finding that GW182 prevents miRNA degradation is potentially interesting, both referees 1 and 2 also point out that, as it stands, the manuscript is rather preliminary and requires significant strengthening before it can be considered for publication by EMBO reports. Both referees remark that endogenous miRNA levels need to be examined in response to GW182 knockdown, and referee 2 adds that alternative methods should be employed to investigate miRNA levels. Referee 1 further indicates that knockdown efficiencies should be demonstrated for XRN1/2 and TNRC6C, and referee 2 mentions that possible indirect effects of shRNA overexpression on miRNA stability should be excluded, or at least addressed.

Defining a new role of GW182 in maintaining miRNA stability.

GW182 binds to Argonaute (AGO) proteins and has a central role in miRNA-mediated gene silencing. Using lentiviral shRNA-induced GW182 knockdown in HEK293 cells, this study identifies a new role of GW182 in regulating miRNA stability. Stably knocking down GW182 or its paralogue TNRC6B reduces transfected miRNA-mimic half-lives. Replenishment of GW182 family proteins, as well as one of its domain...

mRNA degradation by miRNAs and GW182 requires both CCR4:NOT deadenylase and DCP1:DCP2 decapping complexes.

MicroRNAs (miRNAs) silence the expression of target genes post-transcriptionally. Their function is mediated by the Argonaute proteins (AGOs), which colocalize to P-bodies with mRNA degradation enzymes. Mammalian P-bodies are also marked by the GW182 protein, which interacts with the AGOs and is required for miRNA function. We show that depletion of GW182 leads to changes in mRNA expression pro...

The GW/WG repeats of Drosophila GW182 function as effector motifs for miRNA-mediated repression

The control of messenger RNA (mRNA) function by micro RNAs (miRNAs) in animal cells requires the GW182 protein. GW182 is recruited to the miRNA repression complex via interaction with Argonaute protein, and functions downstream to repress protein synthesis. Interaction with Argonaute is mediated by GW/WG repeats, which are conserved in many Argonaute-binding proteins involved in RNA interferenc...

Our Journey from the Study of Human Autoantibodies to the microRNA World

Our discovery of a novel cytoplasmic domain in HEp-2 cells recognized by the autoimmune serum of a patient led us on a journey into the world of microRNAs (miRNA). Our journey began with the identification and cloning of the novel glycine (G)–tryptophan (W)-rich protein, GW182, and its close interaction with the miRNA-binding protein, argonaute2 (Ago2). Discovering the important relevance of GW...

The GW182 protein family in animal cells: new insights into domains required for miRNA-mediated gene silencing.

GW182 family proteins interact directly with Argonaute proteins and are required for miRNA-mediated gene silencing in animal cells. The domains of the GW182 proteins have recently been studied to determine their role in silencing. These studies revealed that the middle and C-terminal regions function as an autonomous domain with a repressive function that is independent of both the interaction ...
